When running Gentoo Linux on my eVGA X58 board, I used to make a 4 GB RAM drive and move the \tmp partition (I think it was) to the RAM drive, which massively sped up the compilation of apps (since Gentoo can be built as a LFS from start to finish, which is how I did it). That machine had 12 GB of RAM.
My current rig has 128 GB RAM - I could probably benefit from one, even as large as 32 GB, without any real ill effect, but subjectively, right now not much feels slow, so I'm not sure of any real subjective benefit.
If games didn't take up so much space, though, I'd try doing it with a game - that would be wild, loading files from RAM to RAM instead from drive to RAM lol.
